BindingSolution Platform Uses High Flexibility and Robustness of Octet® RH16 and RED96e to Address the Characterization of Biomolecular Interactions 

Sartorius Octet® RH16 and RED96e systems offer the best performance for quantitation and kinetic analysis, with the broadest dynamic range and highest sensitivity. These instruments have the ability to measure weak, tight and fast binding interactions with molecular size of analyte from 150 Da to 1000 KDa.

Octet® RH16 system offers biomolecular interactions analysis with state-of-the-art high throughput platform that speeds up library screening, hit and lead discovery and characterization. RH16 has also made it a favorite for compound quantitation in complex mixtures and unpurified samples such as cell culture supernatants and lysates. Its robustness enables to analyse binding kinetic of several dozen of compounds or to screen several hundred of biomolecules over day. But most of all, it is perfectly suitable for highly complex binding matrix such as epitope binning or epitope mapping.

Its scalable throughput offers the possibility to simultaneously analyze up to 16 samples in parallel in a 96 or 384-wells plate format and enables to perform comparative kinetics experiments side by side (e.g binding kinetic at 2 pH conditions, allosteric effect in the presence or not of cofactor…).

Octet® RED96e for its part offers less sample analysis throughput (up to 8 channels simultaneously used on a 96- wells plate), but it is fully compatible for long experimental run times (e.g protein or virus particle quantitation, fast Koff screening, slow koff kinetic measurements…). An evaporation cover facilitates up to 12 hours of unattended run time, while minimizing losses in sample volume and allowing post-analysis sample recovery at the end of the experiment. The sample plate temperature can be controlled from 15 to 40°C, enabling kinetic determinations at different temperatures for temperature-sensitive proteins. Additional advantages of sample cooling include the ability to rapidly determine binding rate constants at multiple temperatures to extrapolate thermodynamic measurements.

Both Octet® devices enable biosensors loaded with ligand to return if required to the biosensor tray at the end of the experiment for additional uses. 

Ligand or analyte consumptions are in the sub-nanomole quantity range in 200 µL final volume for RED96e, down to 50 ul for RH16. Besides, there is still the possibility to re-use same ligand across all the plate and conversely to re-use same analyte for different ligands interaction. Importantly, if required, the samples (ligand and analyte) or standards for quantitation can be fully recovered after the run
